ABSTRAK
Di sekitar Jalan dr Mansur, Medan terdapat cukup banyak rumah makan
yang menyediakan pelbagai jenis makanan dan minuman. Dengan banyak faktorfaktor bakteri untuk hidup dan berkembang biak pada air kran kita, terdapat
kemungkinan besar air kran di rumah makan ini tercemar bakteri coliform.
Bakteri coliform termasuk Escherichia coli merupakan indikator adanya polusi
kotoran dan kondisi yang tidak baik terhadap air minum. Konsumsi air yang
tercemar ini dapat memudaratkan kesehatan manusia.
Penelitian ini bertujuan untuk mengamati kualitas air kran di rumah makan
di Jln Dr. Mansur melalui indikator-indikator mikrobiologi.
Jenis penelitian ini bersifat deskriptif observasional dengan pendekatan
cross sectional. Kedua belas sampel diambil dari rumah makan yang berbeda di
sekitar Jalan dr Mansur,Medan kemudian diperiksa di Laboratorium Mikrobiologi
Fakultas Kedokteran Universitas Sumatera Utara. Penelitian ini menggunakan uji
Most Probable Number (MPN) untuk menghitung jumlah koloni bakteri coliform
terdiri dari uji pendugaan dengan media Lactose Broth (LB), dilanjutkan dengan
uji penegasan dengan media Brilliant Green Lactose Broth (BGLB) 2%. Untuk
melihat ada tidaknya Escherichia coli dilanjutkan uji pelengkap dengan
menggunakan media agar Eosin Methylene Blue (EMB).
Dari hasil penelitian yang dilakukan terhadap dua belas sampel air kran
didapatkan sembilan sampel air kran (75 %) memiliki jumlah koloni bakteri
coliform dengan indeks Most Probable Number (MPN) lebih dari 0 per 100 ml.
Tiga sampel lainnya (25%) tidak terdapat bakteri colifrom dengan indeks MPN 0
per 100 ml. Dari sembilan sampel air kran tersebut, lima sampel positif
didapatkan Escherichia coli pada media agar EMB.
Dapat disimpulkan bahwa air kran di rumah makan di sekitar Jalan Dr
Mansur,Medan mengandung bakteri coliform khususnya Escherichia coli dan air
kran dari rumah makan ini tidak memnuhi syarat kesehatan air minum secara
bakteriologis .

ABSTRACT
There are many restaurant that sell food and drinks around the Jalan dr
Mansur area. Taking into account that there are many factors that contribute to
the growth and cultivation of bacteria in the tap water, thus there is a high
possibility of the water being poluted by bacteria. Coliform bacteria including
Escherichia coli is an indicator of water polution by waste and is not healthy to
consume bu human. Poluted water may be hazardous to human health and well
being.
The aim of this study is to determine the quality of tap water at the
restaurants around Jalan dr Mansur using microbiological indicators and the
examination was carried out at the North Sumatera Faculty of Medicine’s
Laboratory of Microbiology. This study uses the test of Most Probable Number
(MPN) to calculate the number of colonies of Coliform bacteria. This consists of
presumptive test using Lactose Broth (LB) media, followed by a confirmative test
with Brilliant Green Lactose Broth (BGBL) 2% media. The presence or absence
of Escherichia coli was then viewed by a completed test using Eosin Methyl Blue
(EMB) media.
From the examination carried out on twelve samples of tap water
obtained, nine samples (75%) had a colony count of Coliform bacteria with MPN
index greater than 0 per 100 ml. Three other samples (25%) contained no
Coliform bacteria with MPN index 0 per 100 ml. From the nine samples of water,
there are five samples that were positive for the presence of Escherichia coli on
EMB media.
It is concluded that the tap water in food shops in the area Jalan Dr
Mansur, Medan contained Coliform bacteria, especially Escherichia coli and does
not fulfil the bacteriological requirements of healthy drinking water to be
consumed.
