UJI PENURUNAN KADAR KOLESTEROL DARAH MARMOT JANTAN (Cavia porcellus) DARI EKSTRAK DAUN JATI BELANDA (Guazuma ulmifolia Lamk) ABSTRAK Kadar kolesterol tinggi merupakan salah satu faktor resiko utama penyebab arterosklerosis. Pengobatan untuk penderita kolesterol tinggi dengan obat-obatan kimia harganya relatif mahal dan memiliki efek samping sehingga masyarakat sudah menggunakan obat tradisional untuk menurunkan kadar kolesterol yang tinggi. Banyak tumbuhan obat yang digunakan, salah satunya adalah daun Jati Belanda (Guazuma ulmifolia Lamk) suku Sterculiaceae. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak daun Jati Belanda terhadap kadar kolesterol darah marmot yang dibuat hiperkolesterolemia dengan memberikan makanan induksi berupa pakan yang dicampur dengan kuning telur (dosis 1% BB) serta lemak kambing 15 g/100 g jumlah pakan selama 14 hari. Pada penelitian ini dilakukan karakterisasi simplisia, ekstraksi serbuk simplisia dengan cara perkolasi dan uji efek penurunan kadar kolesterol ekstrak daun Jati Belanda terhadap marmot jantan yang dibuat hiperkolesterolemia. Ekstrak daun Jati Belanda diberikan secara oral dengan dosis 50, 100, 200, dan 400 mg/kg BB. Simvastatin digunakan sebagai kontrol positif dengan dosis 0,80 mg/kg BB, dan suspensi Na-CMC 0,5 % sebagai kontrol negatif. Hasil pemeriksaan karakterisasi simplisia daun Jati Belanda diperoleh kadar air 9,90 %, kadar abu total 9,77 %, kadar abu tidak larut dalam asam 0,37 %, kadar sari larut dalam air 25,29 %, kadar sari larut dalam etanol 15,56 %. Pemberian ekstrak daun Jati Belanda dengan dosis 100 mg/kg BB memberikan persentase penurunan kadar kolesterol yang paling tinggi dan merupakan dosis yang paling efektif serta lebih kuat dibandingkan dengan simvastatin dosis 0,8 mg/kg BB. Universitas Sumatera Utara THE TEST IN LOWERING BLOOD CHOLESTEROL LEVEL OF MALE GUINEA PIG (Cavia Porcellus) FROM BASTARD CEDAR LEAVES EXTRACT (Guazoma Ulmifolia Lamk) ABSTRACT High cholesterol levels is one of the major risk factors causing atherosclerosis. The treatment for high cholesterol sufferers with chemical drugs is relatively expensive and have side effects so that the community has been using traditional medicine for lowering high cholesterol levels. Many medicinal plants are used, and one of them is bastard cedar’s leaves (Guazuma ulmifolia Lamk) Sterculiaceae family. The aim of this study was to determine the effect of bastard cedar leaves extract against guinea pig blood cholesterol levels which are made hypercholesterolemic by providing food induction of feed mixed with egg yolk (a dose of 1% BW) and goat fat 15 g / 100 g of food during 14 days. In this research, characterization of simplex has done, extraction simplex powder by using percolation method and test activity of cholesterol-lowering effect bastard cedar extract to male guinea pig that induced hypercholesterolemia were done. Bastard cedar extract was administered orally at a dose of 50, 100, 200, and 400 mg /kg BW. Simvastatin used as a positive control with a dose of 0.80 mg / BW, and the suspension of Na-CMC 0.5% as a negative control. The result of the characterization of crude drug obtained bastard cedar leaves water content of 9.90%, total ash 9.77%, ash insoluble in acid 0.37%, levels of water soluble extract 25.29%, grade 3 in ethanol-soluble extract 15.56%. The results of blood cholesterol lowering activity of guinea pig showed that extract of bastard cedar leaves at dose of 100 mg /kg BW gave the highest a reduction in cholesterol levels and have effective dose and more stronger than simvastatin at dose 0.80 mg /kg BW.
