IDENTITAS PENULIS JUDUL Pengaruh Ekstrak Cumi-Cumi (Loligo sp) Terhadap Kadar TNF-α dan TGF-β1 Serum pada Penyembuhan Luka Tikus Model Diabetes Mellitus The Effect of Squid Extract (Loligo sp) Toward The Levels of TNF-α and TGF-β1 Serum for Wound Healing of a Rat Diabetes Mellitus Model ABSTRAK Diabetes Mellitus (DM) adalah suatu penyakit kronis yang ditandai oleh peningkatan kadar glukosa darah (hiperglikemia) dan dikarenakan oleh resistensi insulin atau defisiensi insulin, sehingga menyebabkan terjadinya lambatnya proses penyembuhan luka. Produksi sitokin proinflamasi yang berlebihan, penurunan TGF-β1, peningkatan jumlah makrofag dan neutrofil dapat menjadi salah satu karakteristik adanya keterlambatan penyembuhan luka. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak cumi-cumi terhadap kadar TNF-α serum, kadar TGF-β1 serum, jumlah makrofag, jumlah neutrofil, dan eritema. Penelitian menggunakan metode Randomized Posttest Only Controlled Group Design dilakukan terhadap tikus Galur Wistar. Hewan coba dibagi menjadi 6 kelompok berupa luka normal dan diabetes dengan diet standar, pemberian suplemen chitosan, ekstrak cumi-cumi secara oral satu kali sehari, dua kali sehari, dan dua hari sekali. Kadar TNFα dan TGFβ 1 diamati melalui metode ELISA, makrofag dan neutrofil menggunakan pewarnaan H&E, eritema dihitung menggunakan program Corel Photo-Paint X6, dan dianalisis statistika dengan menggunakan uji One-Way ANOVA. Hasil penelitian menunjukkan bahwa ekstrak cumi-cumi efektif dalam menurunkan kadar TNF-α serum, jumlah makrofag, dan jumlah neutrofil. Namun tidak mempengaruhi penurunan eritema dan peningkatan kadar TGF-β1 serum. Kata kunci: luka kondisi diabetes, TNF-α, TGF-β1, cumi-cumi ABSTRACT Diabetes Mellitus (DM) is a chronic disease characterized by elevated levels of blood glucose (hyperglycemia) due to impaired insulin action in glucose metabolism resulting in impaired wound healing. Excessive production of pro-inflammatory cytokines, increased number of macrophages and neutrophils, and decrease levels of TGFβ1 serum can be the characteristics of impaired wound healing. This study aims to determine the effect of squid extract to some wound parameters such as levels of TNF-α, and TGF- β1 serum, number of macrophages and neutrophils, and erythema. The research designed as Posttest Only Randomized Controlled Group Design conducted on experimentally male Wistar rats. Experimental animals were divided into 6 groups including normal and diabetic wound with standard diet, diabetic wound with chitosan supplement, diabetic wound given orally squid extract once a day, twice a day, and once in two days. Levels of TNF-α and TGF-β1 serum were observed by ELISA, macrophages and neutrophils by H & E staining, erythema was calculated using Corel Photo-Paint program X6, and analyzed statistically by One-Way ANOVA. The results indicated that squid extract is effective in lowering levels of TNF-α, serum the number of macrophages and neutrophils. However, it has insignificantly shown reduction of erythema and increasing levels of TGF-β1 serum. Keywords: diabetic wound, TNF-α, TGF-β1, squid Author’s Identity: 1.
