PENGARUH SPERMATOZOA BANGSA SAPI YANG BERBEDA TERHADAP PERKEMBANGAN EMBRIO IN VITRO Nang Pranawa Adyaksa 13/359695/PPT/000856 INTISARI Penelitian ini bertujuan untuk mengetahui pengaruh spermatozoa bangsa sapi Simmental, Wagyu, Brahman dan Bali terhadap perkembangan embrio in vitro. Ovarium diambil dari rumah potong hewan (RPH), dibawa dalam larutan NaCl pada suhu 31 sampai 34°C ke laboratorium BET. Oosit dibagi ke dalam empat kelompok berdasarkan spermatozoa bangsa sapi yang digunakan untuk proses fertilisasi. Selanjutnya oosit diinkubasi selama 22 jam untuk proses maturasi. Sperma beku dithawing kemudian dicuci lalu dimasukkan dalam centrifuge tube, dicuci 2 kali. Sperma diinkubasi selama 5 jam untuk kapasitasi. Oosit yang telah dimaturasi dicuci dan diinkubasi dalam drop semen untuk proses fertilisasi selama 5 jam. Oosit yang telah difertilisasi, selanjutnya dicuci dengan medium kultur (CR1aa) dan diinkubasi. Pengamatan perkembangan embrio dilakukan pada hari ke-6 hingga hari ke-8. Data evaluasi embrio yang telah diperoleh dianalisis secara deskriptif. Pengaruh sperma dari bangsa sapi yang berbeda terhadap perkembangan embrio in vitro dianalisis dengan oneway ANOVA. Hasil dalam penelitian menunjukan spermatozoa dari bangsa sapi yang berbeda berpengaruh terhadap perkembangan embrio secara in vitro yaitu Simmental (26,15 ± 7,579), Bali (41,21 ± 9,076), Brahman (40,74 ± 6,249), dan Wagyu (32,19 ± 2,867). Perkembangan embrio terbaik diperoleh dengan menggunakan spematozoa sapi Bali. Fertilisasi in vitro dengan menggunakan spermatozoa sapi Bali dan sapi Wagyu menghasilkan kualitas embrio rata-rata pada grade III, sedangkan sapi Simental dan sapi Brahman menghasilkan kualitas embrio rata-rata pada grade IV. Kata kunci: Spermatozoa, Bangsa sapi, Fertilisasi in vitro, Perkembangan embrio. xii THE EFFECT OF DIFFERENT BREED CATTLE SPERMATOZOA ON THE IN VITRO EMBRYO DEVELOPMENT Nang Pranawa Adyaksa 13/359695/PPT/000856 ABSTRACT This research was conducted to determine the effect of Simmental, Bali, Brahman, and Wagyu spermatozoa on the in vitro embryo development. Ovaries were collected from slaughter house and transportation into sodium chloride (NaCl) solution at 31 to 34oC to BET’s laboratory. Oocytes were collected from ovaries by aspiration method. Oocytes were incubated during 22 hour for maturation. Oocytes were divided into four groups base on the kind of bull breed spermatozoa. Frozen semen were thawed, flushed, and centrifuged five minutes for washing. The semen were incubated during 5 hours for capacitazion. Matured ocytes were flushed using oocyte flushing medium and then incubated for 5 hours in the semen drop for fertilization process. Fertilized oocytes were flushed and incubated for two day. The embryo development was observed from day sixth to eighth post fertilized. The data of embryo development were analyzed descriptive. The effect of the different breed cattle spermatozoa on embryo development analyzed using oneway ANOVA. The result showed that the spermatozoa from different breeds had affected on vitro embryo development: Simmental (26.15 ± 7.579); Bali (41.21 ± 9.076); Brahman (40.74 ± 6.249), and Wagyu (32.19 ± 2.867). The best result of embryo development was showed by Bali breed. In vitro fertilization using Bali and Wagyu sperm given the embryo quality at the third grade, while Simmental and Brahman at the fourth grade.
